Effect of anti-ouabain antibodies on AMPH-induced hyperactivity in mice. Male BALB/c mice were administered saline (10 mL/kg IP) and nonspecific IgG (1 μg/kg ICV) (Control) or saline and anti-ouabain antibodies (1 μg/kg ICV) (Anti-Ou Ab) or AMPH (5 mg/kg IP) and IgG (AMPH) or AMPH and anti-ouabain antibodies (AMPH + Anti-Ou Ab) and tested in the open field test. Three parameters were determined: (A) the distance moved (one-way ANOVA: F3,20 = 7.24; p = 0.002), (B) the duration of mobility (one-way ANOVA: F3,20 = 7.02; p = 0.002) and (C) the mean velocity (one-way ANOVA: F3,20 = 7.16; p = 0.002). n = 8 mice per group. Bars represent means; error bars represent standard error of the means; * different from the Control group; # different from the AMPH group (Holm-Sidak test post hoc; p < 0.05). These results demonstrate that reduction in endogenous cardiac steroids (by anti-ouabain antibodies) affects behavior (reduction in Amphetamine-induced hyperactivity).

Effect of a combination of MK-2206 and ouabain on force of contraction following myocardial infarction (MI) in rats. Left anterior descending coronary artery (LAD) ligation was performed in male Wistar rats. The control group (Ctrl) received an IP injection of saline (0.5 mL·kg−1·d−1), ouabain (0.8-8 mg·kg−1·d−1), MK-2206 (12 mg·kg−1·d−1, MK-2206), or a combination of MK-2206 and ouabain (Oua + MK). Echocardiography was used to monitor heart contractility in vivo. Differences between the fractional area change (ΔFAC) before LAD ligation and the experimental conditions are depicted. Results are expressed as means ± SEM from 2 independent experiments of 5 to 7 animals in each. #Significantly lower than baseline level (P < .05). *Significantly higher than 24 hours. Post LAD ligation level (P < .05). These results show the beneficial effects of the combined treatment with cardiac steroids and AKT inhibitors.

Effect of amphetamine and anti-ouabain antibodies on ERK and Akt phosphorylation levels in the frontal cortex. Male BALB/c mice were administered saline (10 mL/kg IP) and nonspecific IgG (1 µg/kg ICV) (Control, n = 10), saline and anti-ouabain antibodies (1 µg/kg i.c.v.) (Anti-Ou Ab, n = 10) or AMPH (5 mg/kg IP) and IgG (AMPH, n = 10) or AMPH and anti-ouabain antibodies (AMPH + Anti-Ou Ab, n = 10). The mice were sacrificed and the protein levels were determined by Western blot analysis. The values are presented as the mean ± SE (error bars). * p < 0.05. These results demonstrate that the reduction in endogenous cardiac steroids attenuates the increase in ERK and AKT phosphorylation induced by Amphetamine.
